  • Tris(hydroxymethyl)aminomethane( 77-86-1)
Tris(hydroxymethyl)aminomethane( 77-86-1)

Tris(hydroxymethyl)aminomethane( 77-86-1)

Overview: The Chinese name of Tris is trimethylolaminomethane; Aminobutyric triol; Bradyamine; 2-Amino-2 - (hydroxymethyl) - 1,3-propanediol. It is a white crystal or powder. Soluble in ethanol and water, slightly soluble in ethyl acetate and benzene, insoluble in ether and carbon tetrachloride, corrosive to copper and aluminum, irritant chemicals

Application: Trimethylolaminomethane is widely used in acute metabolic and respiratory acidemia. It is an alkaline buffer and has a good buffer effect on metabolic acidosis and enzyme activity reaction. Tris is often used as biological buffer, and is often prepared with pH values of 6.8, 7.4, 8.0, and 8.8. Its structural formula and pH value vary greatly with temperature. Generally speaking, the PH value decreases by 0.03 for each degree of temperature rise. Tris is widely used in the preparation of buffer solutions in biochemistry and molecular biology experiments. For example, the commonly used TAE and TBE buffer solutions (used for nucleic acid dissolution) in biochemistry experiments need to use Tris; Because it contains amino groups, it can be condensed with aldehydes. The pH of the aqueous solution of Tris base is about 10.5. Generally, hydrochloric acid is added to adjust the pH value to the required value to obtain the buffer solution with this pH value. However, attention should also be paid to the effect of temperature on pKa of Tris. Because Tris buffer solution is a weak alkaline solution, DNA will be deporotonated in such solution to improve its solubility. People often add EDTA to Tris hydrochloric acid buffer to make "TE buffer", which is used for the stability and storage of DNA. If the acid solution that regulates the pH value is replaced with acetic acid, the "TAE buffer" (Tris/Acetate/EDTA) will be obtained, and if the acid solution is replaced with boric acid, the "TBE buffer" (Tris/Borate/EDTA) will be obtained. These two buffers are usually used in nucleic acid electrophoresis experiments. TAE and TBE prepared by Tris are the most commonly used reagents for DNA electrophoresis. TE (pH8.0) is mainly used to dissolve DNA (TE is called Tris plus EDTA). Tris buffer is not only widely used as solvent for nucleic acid and protein, but also has many important uses; It is used for protein crystal growth under different pH conditions. The low ionic strength of Tris buffer can be used for the formation of intermediate fibers of lamin of C. elegans. At the same time, Tris is also one of the main components of protein electrophoresis buffer. In addition, Tris is also an intermediate for the preparation of surfactants, vulcanization accelerators and some drugs, and it is often used as a titration standard.
